Cardiovascular biomineralization
It has long been known that bone tissue and even marrow can form in human arteries, usually near cholesterol deposits. This process has been attributed to passive crystal precipitation in dying cells. The Cardiovascular Biomineralization Research Group, directed by Dr. Linda Demer, has shown that the process is both regulated and related to embryonic bone formation. They developed the first tissue culture model of vascular calcification, isolated and cloned the cells responsible for producing calcium mineral, and identified the regulatory molecules controlling the process by which the cells differentiate into bone cells. Based on comparisons to bone cells, they are providing evidence that high cholesterol may contribute not only to atherosclerotic calcification, but also to osteoporosis.

In embryogenesis, immature mesenchymal cells aggregate and organize into patterned tissues. Later in life, a pathological recapitulation of this process takes place in atherosclerotic lesions, when vascular mesenchymal cells organize into trabecular bone, cartilage and fat tissue within the artery wall. In the Cardiovascular Biomineralization Research Group directed by Dr. Linda Demer, a tissue culture model for this process was developed to show that the ectopic tissue is formed by mesenchymal stem cells present in the artery wall that undergo abnormal lineage acquisition. They further showed that these cells self-organize in vitro into patterns that are predicted by a mathematical model based on molecular morphogens interacting in a reaction-diffusion process governed by partial differential equations. They identified the activator and inhibitor morphogens as bone morphogenetic protein and its inhibitor, matrix GLA protein (MGP). The mathematical model predictions were confirmed in culture, and showed that addition of MGP alters the pattern from stripes to spots and that addition of warfarin, an inhibitor of MGP, creates a doubling of stripe density. The stem cells have been grown on carbon nanotubes and nano pillars to attempt to study their left-right chirality and contractile behavior in pattern formation.
