Custom built state-of-the-art super-resolution Stimulated Emission Depletion (STED) microscope: application to basic science in health related research. We built the first custom-made STED microscope in the US which can achieve a resolution of ~30 nm in biological samples. This microscope has the uniqueness of using a resonant scanning mirror (16,000 lines/s) to image a large field of 40 x 40 µm with a pixel size of 10 nm and acquisition speed of 12.5 ns/pixel. This unmatchable resolution of our STED is currently used in studying protein localization and distribution in isolated organelles, cells and tissues This new microscopy was the basis for the creation of the Super Resolution Imaging Center in the Division of Molecular Medicine in the Department of Anesthesiology. At present, we are working towards to: i. Further increase the resolution of STED microscopy by combining our fast scanning STED microscope with other super-resolution methods. ii. To build a two color STED to measure distances with nanoscale resolution between proteins in the same macromolecular complex. iii. To apply this microscopy to basic science in health related research. iiii. To use this microscopy as a new tool for better and more precise pathological diagnostic procedures. 2. Super resolution protein Atlas Initiative: heart, vascular smooth muscles, vascular endothelium and lung 3. macromolecular complexes in a pressure-overload model of cardiovascular disease 4. mechanisms of rapid estrogen induced cardioprotection 5. changes of protein signaling complexes associated with mitochondria during cardioprotection and ischemic preconditioning 6. Additional Information:

Dr. Stefani obtained his MD at the University of Buenos Aires, Argentine and his PhD in Physiology at the Department of Biophysics, University College London under the direction of Dr. R. Miledi FRS and Dr. Sir Bernard Katz, FRS. Dr. Stefani is distinguished by opening new research fields with technical innovations using biophysical approaches. He discovered neuronal serotonin receptors, nerve trophic influence on channel expression, L-type calcium channels in skeletal muscle, and hormonal regulation of ion channels and mechanisms in heart hypertrophy. He pioneered molecular aspects K and Ca channels voltage-sensing mechanisms. Dr Stefani discovered that L-type calcium channels of skeletal muscle as nifedipine receptors can have a role on excitation-contraction coupling. He pioneered studies on voltage dependent ion channel excitability with high resolution recordings in expressed K and Ca channels. He recorded the electrical manifestation of voltage dependent conformation al changes of ion channels referred as gating currents and demonstrated that these precedes pore opening that is also regulated by beta subunits. Recently Dr. Stefani is investigating mechanisms of heart protection during ischemia developing high resolution microscopy techniques beyond the diffraction limit. Dr. Stefani has more than 200 publications in frontier journals
